Yes, this question is sort of specific. I want to be able to access my Ubuntu box at work on my windows machine. I have Putty and have seen examples of using GUI applications through it. I've followed the instructions on different pages, but I just can't get it to work.

You'll need to have an X-Server installed on yer winbox for this to work. If you use Cygwin, you can just use Cygwin's X server. Otherwise I'm sure there are various fine solutions for Windows X serving.
Once you have that running, load the putty session you use to connect to that server, then configure it to do secure X display.
Should work. That's what I do.

X Forwarding is designed really for individual applications, though. Don't try to forward a whole GNOME desktop through it. It can work, but it will be slow. Just start up the programs you want from the command line.
